急性髓性白血病的多药耐药基因-1 C3435T基因多态性与P-gp表达的相关性研究
The association of MDR1 C3435T polymorphism with P-gp expression in acute myeloid leukemia
【摘要】 目的:研究急性髓性白血病(acute myeloid leukaemia,AML)患者的多药耐药基因(multidrug resistance,MDR)-1C3435T基因多态性和P-gp(permeability-glycoprotein)蛋白表达的相关性。方法:采用流式细胞仪测定P-gp表达水平,限制性内切酶多型性—聚合酶链反应测定MDR-1C3435T多态性,并统计其相关性。结果:中国AML患者中MDR-13435基因座上的多态性有自己的分布特点,MDR-13435TT和低P-gp表达显著相关。结论:黄种人AML患者中MDR-13435基因座上的多态性有与白种人不同的分布特点,可能为不同种族的AML患者对相同的化疗药物有效率却不同的原因之一;MDR-13435TT有望预测AML患者化疗敏感程度较其他携带者高,可能可以作为良性预后因素之一。
【Abstract】 Objective To explore the association of MDR1 C3435T polymorphism with P-gp expression in acute myeloid leukemia(AML).Methods P-gp expression was measured by flow cytometry;MDR1 C3435T polymorphism was determined by RFLR-PCR.Their association was examined using Kruskal-Wallis test.ResuLts MDR1 C3435T polymorphism had its unique distribution in Chinese patients with AML.MDR1 3435TT genotype was associated with low P-gp expression.Conclusions The distribution of MDR1 C3435T polymorphism differs between the Chinese patients and the Caucasian patients, which may be one of the reasons for the various responses to the same chemotherapeutic agent in patients of different races.The patients with MDR-1 3435TT genotype have a higher response rate to chemotherapy than those carry any other genotypes.
