真皮多能干细胞对皮肤创面神经修复的影响

【摘要】 目的:观察真皮多能干细胞对皮肤创面神经修复的影响。方法:培养人真皮多能干细胞,以血浆为载体构建移植物;60只裸鼠背部制作皮肤缺损创面,随机分为干细胞移植组(A组)、单纯血浆支架组(B组)和空白组(C组),记录创面愈合时间;创面愈合后第1、2、4、8周取材,免疫荧光检测表皮下及真皮层PGP9.5的表达,以此反映皮肤神经再生情况。结果:A组、B组、C组创面愈合时间分别为(16±1.5)d、(19±2.0)d、(20±1.5)d;A组愈合时间早于B、C组(P<0.05)。A组创面愈合后第1、2周表皮下及真皮层PGP9.5荧光密度高于B、C组(P<0.01);B组第2周真皮层和第4周表皮下PGP9.5荧光密度高于C组(P<0.05)。结论:人真皮多能干细胞复合血浆移植后,在加快皮肤创面愈合的同时可促进神经修复。

【Abstract】 Objective To observe the effects of dermal-derived multipotent stem cells(DMSC) on neural regeneration in skin wounds.Methods DMSC were cultured and seeded on human plasma as transplants.60 naked mice, on whose back full-thickness defect skin wounds were made, were randomly assigned to receive DMSC transplantation(group A), acellular human plasma graft implantation(group B), or no interference(group C).Wound healing time was recorded.The expression of protein gene product 9.5(PGP9.5) in healed skin at week 1, 2, 4, and 8 after wound healing was detected by immunofluorescence.Results Wound healing time in group A was shorter than those in group B and group C(16 ± 1.5) d vs(19 ± 2.0) d and(20 ± 1.5)d, P < 0.05.The fluorescence density value of PGP9.5 in group A was higher than those in group B and group C at week 1 and 2 after wound healing(P < 0.01), and that in group B was higher than that in group C at week 2 and 4(P < 0.05).Conclusions DMSC seeded on human plasma transplanted into skin wounds can accelerate wound healing and neural regeneration.
