HPLC和ELISA法检测食品中呋喃唑酮

【摘要】 目的建立了用于检测食品中呋喃唑酮的高效液相色谱法(HPLC)和酶联免疫吸附试验(ELISA)。方法HPLC法,样品以乙腈提取,正己烷脱脂,SPE(Alumina B)净化,Hypemil ODS C18(4.6×250mm,5μm)进行分离,紫外检测器检测呋喃唑酮。酶联免疫吸附试验(ELISA)法,采用ELISA检测法和酶标读数仪定量检测。结果HPLC法回收率94.8%,相对标准偏差(RSD)为3.4%,呋喃唑酮的最低检测限0.2ng/kg。ELISA法呋喃唑酮含量在50~4050ng/kg之间,精密度CV%为2%~5%,回收率为87.3%~107.5%,标准曲线r=-0.995~-0.999。结论HPLC方法具有灵敏度高、重现性好,但是仪器设备投入高;ELISA法特异性高,干扰少、测定步骤简便、快速,操作安全,设备投资少。

【Abstract】 Objective To detect furazolidone in food by solid phase extraction(SPE)-high performance liquid chromatography and ELISA.Methods The sample was extracted by aeetonitrile,removed ester by hexane,purified by SPE(Alumina B),gradient solvents eluted,separated with a Hypemil ODS C18(4.6×250mm,5um),then detected by HPLC.Quantitative determination was carried out with the advanced ELISA detection method and stat Rayto RT-6100 readout.Results The recovery range of determination was 94.8%,and the relative standard deviation(RSD) was 3.4%.The detection limit of furazolidone was 0.2ng/kg by HPLC.The content of furazolidone was between 50ng/kg and 4,050ng/kg,CV was in the range of 2%~5%,the recovery rate was between 87.3% and 107.5%,and the standard curve r was between-0.995 and-0.999 by ELISA.Conclusions ELISA method is simple,sensitive and rapid,with little interference.HPLC is accurate and reproducible.
