厚朴的提取方法及质量分析考察
Extraction processes and quality analysis of root bark and stem bark of Magnolia officinalis Rehd. et Wils.
【摘要】 目的以厚朴酚、和厚朴酚为含量测定指标,寻找厚朴最为有效的提取方法。方法采用甲醇冷浸法、乙醇热回流法、超临界CO2萃取法(CO2supercritical fluid extraction)对厚朴药材进行提取,利用HPLC法对各提取物中厚朴酚、和厚朴酚的含量进行测定。结果超临界CO2萃取的提取物中厚朴酚、和厚朴酚的含量最高,质量分数分别为2.00%、2.79%,分别是甲醇冷浸法、乙醇热回流法的2.5倍、4倍。结论运用超临界CO2萃取法能对厚朴进行高效率的提取,HPLC法能简便、准确地测定提取物中厚朴酚、和厚朴酚的含量。
【Abstract】 Objective To study the optimum extraction processes of root bark and stem bark of Magnolia officinalis,with magnolol and honokiol as the assessment index.Methods Three methods,which included methanol merceration,ethanol refluxing extract and CO2 supercritical fluid extraction,were applied to extract magnolol,honokiol from Magnolia officinalis.The contents of magnolol and honokiol by the three extraction methods were assayed by HPLC.Results The contents of magnolol,honokiol were the highest by CO2 supercritical fluid extraction(2.00%,2.79% respectively).Conclusions Root bark and stem bark of Magnolia officinalis can be effectively extracted by CO2 supercritical fluid extraction;the HPLC analysis is convenient,easy and suitable to assay the contents of magnolol and honokiol from Magnolia officinalis.
