星形细胞瘤组织CD105与TGF-β1的表达及意义

【摘要】 目的探讨CD105和转化生长因子β1(TGF-β1)在星形细胞瘤中的表达及意义,为星形细胞瘤的生物治疗提供依据。方法采用免疫组化法检测正常脑组织、低级别(Ⅰ~Ⅱ级)和高级别(Ⅲ~Ⅳ级)星形细胞瘤组织及瘤旁组织中CD105和TGF-β1蛋白表达。结果星形细胞瘤组织中CD105标染的微血管密度(CD105-MVD)和TGF-β1蛋白表达水平明显高于瘤旁组织和正常脑组织(P<0.01)。Ⅲ~Ⅳ级星形细胞瘤CD105-MVD和TGF-β1蛋白表达水平均高于Ⅰ~Ⅱ级星形细胞瘤。星形细胞瘤中TGF-β1高表达组CD105-MVD显著高于低表达组和阴性表达组(P<0.05),低表达组高于阴性表达组(P<0.05)。结论CD105与TGF-β1在星形细胞瘤中表达关系密切,可促使肿瘤生长,使其增殖侵袭加速并向恶性转变。CD105与TGF-β1联合检测是判断星形细胞瘤恶性程度的有价值的指标,以此靶点的抗肿瘤治疗将为星形细胞瘤的治疗带来新的选择。

【Abstract】 Objective To study the expression of CD105 and TGF-β1 in human astrocytoma and their significance,and supply evidence for clinical biotherapy.Methods Immunochemical technique was applied to determine the expression of CD105 and TGF-β1 protein in 56 cases with low-grade astrocytoma and paratumor tissue,and 56 cases with high-grade astrocytoma and paratumor tissue,and 10 cases of normal brain tissues were allocated to control group.Results Compared to control group and paratumor tissue,the CD105-MVD and TGF-β1 immunopositive cells were significantly increased in astrocytoma (P<0.01). The expression of CD105-MVD and TGF-β1 were significantly increased in high-grade astrocytoma versus to low-grade astrocytoma. There were statistical significance about CD105-MVD among significantly expressed group,low expressed group and negative group of TGF-β1 (P<0.05).Conclusion The CD105 and TGF-β1 protein are over-expressed in human astrocytoma to a certain extent,and the positive rate of expression is gradually increased as the histological grade becoming more serious,and they might be combined in utilization for discriminating the malignant degree of astrocytoma. So,the new therapeutic regimen for astrocytoma would be provided based on these studies.
