巨细胞病毒特异性IgG抗体检测方法的建立及应用

【摘要】 目的为了建立一种快速、简便、实用的可用于育龄妇女血清中巨细胞病毒(CMV)-IgG抗体的筛查方法,并初步探索其应用价值。方法建立重组CMV(pp150+52)抗原;以国家人口和计划生育委员会科学技术研究所(简称本所)建立的重组CMV(pp150+52)抗原作为包被抗原,700IU/L作为临界值,用间接ELISA方法,对深圳宝安区1024例血清标本进行CMV-IgG抗体的检测。结果该方法灵敏度为13IU/L,批内变异系数4.7%~7.0%,批间变异系数7.6%~14.4%,并与SORIN公司生产的试剂盒进行了比较,2种方法呈最度相关,Kappa值为0.82;深圳宝安区1024例血清标本CMV-IgG抗体阳性率为93.3%(955/1024),阴性率为6.7%(69/1024)。结论本所建立的ELISA方法快速、简便和实用,可用于育龄妇女CMV-IgG抗体的筛查。

【Abstract】 Objective To establish a rapid, serum and convenient method for the screening of cytomegalovirus (CMV) specific IgG antibody in sera samples of women of childbearing age, and to evaluate the preliminary significance of application. Methods The measurement of CMV-IgG was conducted by indirect enzyme linked immunosorbent assay using recombinant CMV-Ag (pp150+52) as coating antigen, and 700 IU/L as Cut-off value. Results The specific CMV-IgG antibodies in sera were detected in 1 024 women of childbearing age in Shenzhen, by using the method established by our study and the ELISA assay kid provided by Diasorin Co. The positive rate, negative rate, sensitivity of our method was 93.3 %, 6.7 %, and 13 IU/L, respectively. The intraassay coefficient of variation was between 4.7 %-7.0 % and the interassay coefficient of variation was between 7.6%-14.4 %. The Kappa value between our method and ELISA assay kid provided by Diasorin Co. was 0.82, indicating a good relativity. Conclusion Our ELISA assay is a rapid, easy and convenient method and could be used for screening the CMV-IgG specific antibodies among premarital women.
