脂质体介导端粒酶反义RNA对人乳腺癌细胞MCF-7活性及增殖的影响
The effect of liposome-mediated telomerase antisense RNA on telomerase activity and cell proliferation of MCF-7 cells
【摘要】 目的:探讨经脂质体介导转染端粒酶反义RNA对MCF-7细胞端粒酶活性及细胞增殖的影响。方法:用脂质体介导法将pBBS212-hTR(反义端粒酶RNA真核表达载体)导入人乳腺癌细胞MCF-7中,采用TRAP-PCR-ELISA法测定端粒酶活性,四唑盐比色(MTT)法检测MCF-7细胞的增殖,FCM分析等方法观察其对MCF-7细胞的增殖影响。结果:端粒酶反义RNA能显著抑制MCF-7细胞的端粒酶活性,抑制MCF-7细胞的增殖并诱导其凋亡。结论:脂质体转染反义端粒酶RNA在封闭细胞端粒酶活性表达的同时,可促进MCF-7细胞凋亡。
【Abstract】 Objective:To investigate the effects of liposome-mediated antisense telomerase RNA on telomerase activity and cell proliferation of MCF-7 cells.Methods:Human breast cancer MCF-7cell line was transfected with antihygromycin antisense telomerase RNA eucaryotic expression vector,pBBS212-hTR,mediated by lipofectamine2000(LF2000).Telomerase activity of MCF-7 cell strain was analysed by TRAP-PCR-ELISA technique.The inhibiting rate was determined by MTT assay,flow cytometry was used to examine cell cycle.Results:Compared with control,cell apoptosis rose significantly in the course of antisense telomerase RNA inhibtion of MCF-7 telomerase activity.Conclusion:Antisense telomerase RNA can promote MCF-7 apoptosis by inhibiting the telomerase activity.
