川芎嗪对血管再狭窄大鼠血小板源生长因子受体与细胞外信号调节蛋白激酶表达的影响
The inhibiting effects of tetramethylpyrazine on vascular restenosis in rats
【摘要】 目的:观察血小板源生长因子(PDGF)受体与细胞外信号调节蛋白激酶(ERK1/2)在再狭窄血管中的表达及川芎嗪(TMP)对其的影响,探讨TMP防治血管再狭窄的作用机制。方法:制备大鼠血管再狭窄模型,采用不同浓度的川芎嗪干预,观察颈总动脉形态学改变,免疫组化法检测颈总动脉PDGF-β受体与ERK1/2的表达。结果:假手术组动脉各层结构正常、清晰,无明显血管平滑肌细胞(VSMC)增殖,颈总动脉损伤大鼠血管平滑肌细胞(VSMC)大量增殖,内膜显著增厚,川芎嗪处理后,血管平滑肌细胞(VSMC)增殖程度降低;与假手术组比较,再狭窄大鼠颈总动脉PDGF-β受体与ERK1/2表达显著增强(P<0.01),中、高剂量川芎嗪明显降低PDGF-β受体及ERK1/2的表达(P<0.01,P<0.05)。结论:TMP抑制血管再狭窄,其机制与抑制血管平滑肌细胞PDGF受体及ERK1/2表达有关。
【Abstract】 Objective:To study the effects of tetramethylpyrazine(TMP)on vascular restenosis in rats.Methods:Restenosis model was established.Treatment with TMP in different dose.The morphologic change,the expression of PDGF-βreceptor and extracellular signal regulated protein kinase 1/2(ERK1/2)were detected.Results:Compared with sham,the expression of PDGF-βand ERK1/2 were obviously increased in operation group(P<0.01),the VSMCs proliferation was inhibited and expression of PDGF-βand ERK1/2 were decreased obviously by TMP.Conclusion:TMP play an important role in cure restenosis through inhibiting the expression of PDGF-β receptor and ERK1/2.
