前列地尔对大鼠肢体缺血再灌注后脊髓神经元细胞凋亡的影响
Effects of PGE1 on apoptosis of spinal cord neural cell after ischemia-reperfusion of hind limbs in rats
【摘要】 目的探讨前列地尔对大鼠肢体缺血再灌注脊髓神经元细胞凋亡的影响。方法成年健康SD大鼠72只,随机分为3组:假手术组(sham组,n=24)、缺血再灌注组(IR组,n=24)、前列地尔组(PGE1组,n=24)。3组大鼠均复制实验性大鼠肢体缺血再灌注损伤模型,采用HE染色观察脊髓L1-5节段的病理学改变,TUNEL法检测脊髓细胞凋亡指数(AI),免疫组化检测脊髓神经元Bcl-2、caspase-3表达的变化。结果与sham组比较,IR组和PGE1组Bcl-2、caspase-3阳性细胞率、AI都显著增加(P<0.01),Bcl-2在IR组内于再灌注12h表达达高峰;caspase-3和AI在IR组内于再灌注24h表达达高峰。IR组和PGE1组caspase-3与AI呈正相关。与IR组比较,PGE1组Bcl-2阳性细胞率增加,caspase-3阳性细胞率和AI减少(P<0.05)。结论前列地尔可能通过上调Bcl-2表达和下调caspase-3表达使脊髓神经元凋亡减轻,从而对大鼠肢体缺血再灌注后脊髓神经元有一定的保护作用。
【Abstract】 Objective To investigate the effects of PGE1 on apoptosis of spinal cord neural cells after ischemia-reperfusion of hind limbs in rats.MethodsSeventy-two SD rats were randomly divided into 3 groups:sham operation group(sham group,n=24),ischemia-reperfusion group(IR group,n=24),PGE1 group(n=24).Model of limb ischemia-reperfusion was established in all rats.The pathological changes of spinal cord were assessed by HE staining.The apoptosis index(AI)of spinal cord neural cells was measured by TUNEL method.The expression of Bcl-2 and caspase-3 was assessed by immunohistochemical method.ResultsThe expression of Bcl-2,caspase-3 and apoptosis index of spinal cord neural cells were significantly higher in IR group and PGE1 group than in sham group(P<0.01).The expression of Bcl-2 peaked after reperfusion for 12 h.The expression of caspase-3 peaked after reperfusion for 24 h,while apoptotic cells were the most.In IR group and PGE1 group,caspase-3 was positively correlated with apoptosis index.The expression of Bcl-2 was significantly higher in PGE1 group than in IR group,while the expression of caspase-3 and AI was significantly lower(P<0.05).ConclusionPGE1 can relieve the spinal cord neural cell apoptosis by up-regulating Bcl-2 expression and down-regulating caspase-3 expression,and then protect spinal cord neuron after hind limb ischemia-reperfusion injury.
