【摘要】 目的分析不同HIV抗体检测试剂检测HIV-1B’和BC重组型膜蛋白抗体的敏感性。方法选择HIV-1B’和BC重组型感染者血浆,用gp41抗原和gp160抗原分别进行亲和层析纯化。将各纯化产物进行系列稀释,用15种HIV抗体ELISA检测试剂以及3种HIV抗原/抗体联合检测试剂进行检测。结果用gp41抗原纯化后的样品只含有抗gp160和gp41的抗体带型,而用gp160抗原纯化后的样品含有抗gp160、gp120和gp41的抗体带型。进口试剂检测不同基因型样品的敏感性低于大部分国产试剂,国产试剂检测不同基因型的样品的敏感性无明显差异,而进口试剂检测B’亚型抗体的能力低于检测BC重组型抗体的能力。结论不同基因型的抗体对HIV抗体检测试剂的敏感性可能有一定的影响。更多还原

【Abstract】 Objective To analyze the sensitivities of various HIV antibody detection kits to antibody against env protein of HIV-1.Methods The plasma samples of patients infected with HIV-1 clades B’ and BC was purified by affinity chromatography with gp41 and gp160 antigens respectively.Each sample was diluted serially and detected by 15 kinds of HIV antibody ELISA kits and 3 kinds of HIV antigen /antibody combined detection kits respectively.Results The samples purified with gp41 antigen showed antibody band patterns against gp160 and gp41,while those with gp160 antigen showed antibody band patterns against gp160,gp120 and gp41.The sensitivities of imported kits to the samples of various genotypes were lower than those of most of domestic kits.No significant differences were observed in domestic kits to the samples of various genotypes.However,the sensitivities of imported kits to the samples of clade B’ was lower than those to clade BC.Conclusion The genotype of antibody might influence the sensitivity of HIV antibody detection kit.更多还原
