深海链霉菌选择性分离及活性菌株16S rRNA聚类分析
Selectively Isolated Deep-sea Streptomyces and Analysised 16S rRNA Phylogenetic Treeof Activity Strains
【摘要】 选择性分离深海链霉菌,检测其抗肿瘤细胞,抗金黄色葡萄球(Staphylococcus aureus ATCC 51650),抗PTP1B和抗caspase-3活性,对活性菌株做165 rRNA聚类分析。用8种选择性培养基分离培养,检测活性和构建活性菌株16S rRNA聚类分析图。共分离到90株链霉菌,其中在RH培养基上分离到的菌株数量和类群最多,占38.9%,次之为GS1和OA培养基,而M5培养基未分离到菌株;共筛选到活性菌株44株,有抗肿瘤活性14株,抗金黄色葡萄球菌22株,抗PTP1B活性19株和抗caspase-3活性2株,其中16株至少有两种活性;16S rRNA聚类分析结果表明44株活性菌株分布在4个clade,分别是S.coelicolor,S.pactum,S.stramineus和S.restomycificus clade,以S.coelicolor和S.restomycificus clade为主,分别占66.0%和11.1%。在常温,常压下可分离培养到大量高活性和活性多相样性的深海键霉菌,RH培养基最适用于分离深海链霉菌。
【Abstract】 The aims of this study were to selectively?isolate streptomyces form the South China Sea 2332 m depth sediment, measure their activities against Human Colon Tumor(SMMC-7721) cells,Staphylococcus aureus ATCC 51650,protein tyrosine phosphatase(PTP1B) and caspase -3,and analysis16S rRNA phylogenetic tree of activity strains.we selectively isolated streptomyces by using the eight media,examined to determine their activities in four high throughput screening models and analysised results of the activity strains 16S rRNA phylogenetic tree which was built in MEGA 4.0 software.The total of ninety streptomyce isolates were obtained.The most numbers and diversities of isolates observed on the isolation RH medium plates,occupied 38.9%.Secondly,the media were GS1 and OA.But no strains were cultured on the M5 medium plates;All of 44 activity strains were 14 the activity against the tumor cell,22 against S.aureus,19 against PTP1B,and only 2 isolates inhibited caspase-3.16 isolates exhibited activities in more than two of the screens;The 16S rRNA phylogenetic tree results showed that the 44 activity strains?were divided into four clades,which were S. coelicolor,S.pactum,S.stramineus and S.restomycificus clade,and these mostly were S.coelicolor and S.restomycificus clade, respectively occupied 66.0%and 11.1%.The receach results indicated that a lot of high?and multifarious activity deep-sea streptomyces could be isolated and cultured in the condition of normal temperature and pressure,and the RH medium was the best to isolate streptomyces.
