HBV前S1蛋白与HepG2细胞膜蛋白结合位点鉴定
Attachment Site of Hepatitis B Virus PreS1 Domain for Protein on HepG2 Membrane
【摘要】 为鉴定HepG2细胞膜蛋白识别HBV包膜蛋白preS1区的位点。通过删除突变的方法构建preS1的不同区域片段与GST融合的重组表达质粒,将表达质粒转入E.coli BL21菌株中原核表达,以生物素标记HepG2细胞膜蛋白,pull down试验分析HepG2细胞膜蛋白识别preS1的位点。结果表明,21~33位氨基酸是HepG2细胞膜蛋白识别preS1的主要位点。通过对HepG2细胞膜蛋白与preS1结合的位点的分析,为进一步研究preS1在HBV早期感染中的作用和HBV包膜蛋白受体打下基础。
【Abstract】 Deleted segments of preS1 were recombined with GST tags and expressed in prokaryotic expressing system. GST pull-down assay was employed to identify which segments is essential for the binding of preS1 to protein on HepG2 membrane,using the precleared lysate of biotinylated HepG2 cells as a source of ligands. The results showed that amino acid residues 21~33 was essential for the binding of preS1 to protein on HepG2 membrane.
