DENV2海南分离株NS1全序列测定和生物信息学分析

【摘要】 目的:对登革病毒2型海南分离株NS1全序列进行测定和生物信息学分析,了解其系统进化和分子流行病学特征。方法:采用RT-PCR法扩增D2-Hainan全长NS1基因,将其克隆入载体pPIcZαB,测序,采用相关软件进行生物信息学分析。结果:获得D2-Hainan株NS1全长基因1056bp,其序列与登革病毒2型NGC株的同源性为95%。系统进化树分析显示该毒株与登革病毒2型China04株的亲缘关系最近。初步分析了NS1蛋白的氨基酸序列特征和二级结构。结论:登革病毒流行株存在地域性和复杂性,对D2-Hainan株NS1基因及其编码蛋白信息特征的了解,有助于进一步研究其基因特征与病毒毒力的关系。

【Abstract】 Objective:Determine and analyze the entire sequence of the NS1 gene of dengue 2 virus strain isolated in Hainan which is to understand the characteristic of its molecular evolution.Method:The nucleotides of NS1 of D2-Hainan was amplified by RT-PCR and the sequence was determined by Sanger method.Then the gene and the encoding protein were analyzed by BLAST,MEGA,Clustal X,nnPredict,HNN,and DAS software.Result: It showed that the whole nucleotide sequence of D2-Hainan NS1 was 1 056bp and had the 95% homology with that of NGC strain.It was most closely to China04 strain with the homology 99.3%.Bioinformatics analysis proved there were 352 deduced amino acids of NS1 protein and it shared the twelve highly conserved cysteine sequence among flaviviruses.Conclusion:By carrying out sequencing and bioinformatics of NS1,our research established foundation of further explore for the relationship between the NS1 gene character and dengue virus virulence.
