五氯酚对斑马鱼胚胎发育期Wnt信号通路的影响
Effects of Pentachlorophenol on the Wnt Signaling during the Development of Zebrafish Embryo
【摘要】 为从毒理基因组学角度探讨五氯酚(PCP)发育毒性的作用机制,将0hpf斑马鱼(Danio rerio)胚胎暴露于50μg·L-1PCP中8h,提取其总RNA,与Affymetrix公司的斑马鱼寡核苷酸芯片杂交.结果发现,与对照组相比,染毒样本中,1149个基因的表达显著增强(log2ratio>1),501个基因的表达显著减弱(log2ratio<-1).利用生物信息学软件GenMAPP对差异表达基因进行了Wnt信号通路分析,结果显示,PCP暴露后斑马鱼胚胎发育过程中fzd2、axin2等Wnt信号通路重要的调控基因表达均发生了显著变化;PCP影响了经典Wnt通路和非经典Wnt通路,并可能影响了Wnt通路与FGF通路的相互作用.
【Abstract】 In order to explore the mechanism of development toxicity of pentachlorophenol(PCP)at the point of genomics, 0hpf zebrafish (Danio rerio) embryos were exposed to 50μg·L-1 PCP for 8h in vitro. Total RNA sample was extracted then and hybridized with Affymetrix Zebrafish Genome Array representing approximately 14,900 transcripts. Results showed that a total of 1149 transcripts were significantly up -regulated (log2 ratio>1), while 501 transcripts were down -regulated (log2 ratio<-1). Bioinformatic tools were used for further analysis. Results indicated that genes including fzd2, axin2 that were regulated by Wnt signaling might be involved in the developmental toxicity of PCP. PCP might also affected the interaction of Wnt signaling and FGF signaling.
