多黏性芽孢杆菌HT16抗真菌物质的分离及特性研究

【摘要】 多黏性芽孢杆菌HT16(Paenibacillus polymyx)产生的抗菌物质对扩展青霉菌、桔青霉菌、黑根霉菌等多种病原真菌具有良好的抑制作用。将发酵上清液经过酸沉淀,甲醇抽提得到粗提物,研究显示该粗提物有很好的热稳定性,耐紫外线,对蛋白酶不敏感,耐酸不耐碱,pH在6.5~8.0活性最高,在250nm有最大吸收峰。将粗提物经DEAE-SephadexA50层析,收集活性峰121℃加热,再经10ku超滤膜过滤,电泳检测其为一小肽,分子量小于5ku。抑菌实验表明,孢子萌发率低,菌丝扭曲膨大,分生孢子梗发育不良。

【Abstract】 The antifungal substance produced by Paenibacillus polymyx HT16 has strong inhibition to many pathogenic fungi such as Penicillium expansum, P. cirtrimum and Rhizopus nigricans. Crude extraction of antifungal substance was obtained by precipitation of the cell free supernatant of HT16 with acidic methanol. The study shows that the crude extraction is very stable under high temperature, ultraviolet, proteinase and acid treatments, but not stable under alkali circumstances. Its optimum pH range for anti-pathogenic fungi is between 6.5~8.0. At 250 nm there is a maximal ultraviolet absorption. The crude extraction was further purified by DEAE-sephadex A50 chromatography, the active component was collected and heated at 121 ℃ for 20 min, and then after ultra-filtration, an active component with molecular mass around 5 ku identified by Trcine- SDS-PAGE was obtained. The active substance decreases spore germination rate, causes the swollen and twisted hyphal growth and abnormal conidiophores growth.
