RP-HPLC测定牛膝中齐墩果酸的含量

【摘要】 目的:建立牛膝中齐墩果酸含量的RP-HPLC测定方法。方法:采用超声提取方法;色谱条件:Allsphere ODS-2柱(250mm×4.6mm,5μ),流动相为甲醇-水-醋酸(80∶20∶0.01,v/v/v),柱温30℃,流速1.0mL/min,检测波长215nm。结果:齐墩果酸在0.01~0.4mg/mL范围内线性关系良好,线性方程为Y=2796327X-31842,r=0.9997,平均回收率为98.96%,RSD=0.21%(n=5),比较了不同产地牛膝中齐墩果酸的含量,其中山东产的含量最高。结论:实验方法简单、准确可靠,为牛膝的质量评价提供了实验依据。

【Abstract】 Objective:To establish an RP -HPLC method to determine the content of oleanolic acid in Achyranthes bientata B1.. Methods: By ultrasonic extraction technique, the suitable chromatogram conditions: Allsphere ODS-2 column(250 mm×4.6 mm, 5 μ), the mobile phase was methanol-water-acetic acid(80∶20∶0.01, v/v/v), column temperature was at 30 ℃, flow rate was 1.0 mL/min, UV detection wavelength was 215 nm. Results: The good linear range of Oleanolic Acid was 0.01~0.4 mg/mL, regression equation: Y=2796327X-27842, r=0.9997, the average recovery rate was 98.96%, RSD=0.21%(n=5). The content of Oleanolic acid is the highest in Shandong Province, in comparison of others. Conclusion: The method is simple, accurate and can be used to evaluate the quality of Achyranthes bientata B1.
