蒲公英花黄色素的超声提取工艺及叶黄素含量测定
Ultrasonic extraction of yellow pigment from Dandelion′s petal and quantitative determination of its lutein
【摘要】 从野生蒲公英花中提取的黄色素是一种安全、无毒的天然色素,因其中含有一定量的叶黄素,故其具有很高的营养价值。利用正交设计研究了该色素的超声提取工艺,并且对提取的黄色素用HPLC(高效液相)法进行了叶黄素含量测定。提取最佳条件为提取时间90min、提取温度55℃、提取功率60W;HPLC条件:使用十八烷键合硅胶色谱柱,流动相为乙腈∶甲醇(95∶5),检测波长为446nm。该方法在3.6~18.0μg/mL范围内线性关系良好,加样回收率为100.03%。
【Abstract】 The yellow pigment extracted from Dandelion’s petal is safe and nonpoisonous. To consist of a lot of lutein it is also a nature pigment with certain nutritive vaLue. We studied ultrasonic extraction and quantitative determination of lutein by HPLC. The parameters of optimum extraction process: frequency of ultrasound to extracting is 60 W, extractible temperature is 55 ℃, extractible time is 90 minutes. The HPLC used Diamonsil C18 column, a mixture of (CH3CN∶CH3OH=95∶5) as mobile phase, and UV detection at 446 nm. Lineal relation is better when the sample is 3.6~18.0 μg/mL. The recovery of added lutein is 100.03%.
