不同剂量蛹虫草菌丝体对大鼠免疫功能的影响
EFFECTS OF THE CULTURED MYCELIA OF CORDYCEPS MILITARIS ON IMMUNE FUNCTION IN RATS
【摘要】 研究采食不同水平的蛹虫草菌丝体对大鼠免疫功能的影响。选取60只初始体重(190.6±0.9)g的SD雄性大鼠,随机分为5个处理组,分别饲喂含有0(基础日粮)、0.1%、0.2%、0.5%、1.0%蛹虫草菌丝体的日粮。试验期21d,所有大鼠于14d腹腔注射20%的SRBC0.5mL进行免疫刺激,在21d试验结束之前采血测定淋巴细胞转化率、血清溶血素、血清免疫球蛋白A、M、G以及细胞因子IL-2、IL-4、IL-10、IFN-γ、TNF-α含量,采血后处死大鼠测定胸腺指数和脾脏指数并制取脾细胞,进行脾细胞增殖以及脾定量溶血分光光度测定。结果表明,日粮中添加一定浓度的蛹虫草菌丝体可以显著提高大鼠的胸腺指数、脾定量溶血分光光度以及血清溶血素、IgG、IgM和细胞因子IL-2、IL-4、TNF-α含量,说明日粮中添加蛹虫草菌丝体能够提高大鼠的免疫功能特别是体液免疫水平。
【Abstract】 Effects of the cultured mycelia of Cordyceps militaris (CM) on immune function in rats were investigated. Sixty male SD rats (190.6 ± 0.9) g were fed based diets containing 0,0.1 %,0.2 %,0.5 % or 1.0 % culturedmyceliaof Cordycepsmilitaris respectivelyduringa21dexperiment.Eachratwas injected intraperitoneally with 0.5 mL sheep red blood cells as a immunization. Blood samples were collected on 21 d before all rats were slaughtered to determine the index of immune organs and to collect spleen samples. Lymphocyte proliferation of blood and spleen cells,hemolytic activity (HA),quantitative hemolysis of sheep red blood cells (QHS) and serum concentrations of Ig G,Ig A,Ig M and IL-2,IL-4,IL-10,IFN-γ and TNF-α were evaluated. The results showed that dietary supplement of CM could increase the weight of thymus,QHS,and serum concentrations of Ig G,Ig M,IL-2,IL-4,TNF-α .The results suggested that CM could improve the humoral immune function in rats.
