响应曲面法优化兽疫链球菌变异株透明质酸发酵条件的研究
Optimization of fermentation conditions for the production of hyaluronic acid by Streptococcus zooepidemicus mutant T-07 using response surface method
【摘要】 通过离子束诱变,获得了兽疫链球菌变异株T-07。根据单因素实验,发现影响兽疫链球菌变异株T-07透明质酸(HA)产量的关键因素为时间、温度和葡萄糖浓度。采用三因素三水平Box-Behnken实验设计,应用响应面分析法(RSM)对该变异株发酵生产HA的关键因素条件进行优化。经过优化,HA产量提高了3.0倍,在2.1g/L以上,实验值与预测值基本相符。
【Abstract】 Streptococcus zooepidemicus mutant T-07 was obtained by mutation breeding using low-energy ion implantation.After the single factor experiments,the important and critical culture conditions for production of hyaluronic acid(HA) were set as fermentation time,temperature,and concentration of glucose culture.In addition,response surface methodology(RSM) was used to optimize the critical culture conditions of HA fermentation using a Box-Behnken design(BBD).Through optimization,the yield of HA increased 3.0 times,up to 2.1g/L.
