【摘要】 对食用油中AFB1的含量进行分析,探索建立免疫亲和柱净化、提取液衍生、高效液相色谱结合荧光检测器测定食用油中AFB1的方法。甲醇-乙腈-水(10+30+60),Agilent-C18柱(4.6mm×300mm,5μm),流速为0.6mL/min,激发波长为365nm,发射波长为440nm,进样量为20μL。实验表明:AFB1在10~50ng/mL时,浓度与峰面积呈线性关系(r=0.9936),加标回收率为76.32%~88.97%,最低检出限为0.2μg/kg,相对标准偏差为1.23%~4.62%。该方法测定食用油中AFB1的含量,简便快速、灵敏准确、重复性好。更多还原

【Abstract】 The AFB1 content in edible oils were analyzed in order to develop a method of determination of AFB1 content in edible oils by using immuno affinity columns cleaning up,extraction derivatization,HPLC combining with fluorescence detector.Chromatographic conditions included Agilent-C18 column(4.6mm × 300mm,5μm)and the mobile phase consisting of a mixture of methanol,acetonitrile and water by 10,30 and 60(v/v).The flow rate was 0.6mL/min.Exciting wavelength was at 365nm and emission wavelength was at 440nm.The sample dose was 20μL.The result showed that when the AFB1 content was at 10ng/mL to 50ng/mL,the perfect linearity between concentration and peak area ratio was obtained(r=0.9936),the recovery rate was 76.32% to 88.97%,and the detection limit was 0.2μg/kg,the relative standard deviation(RSD)was 1.23% to 4.62%.This method was simple,rapid,sensitive,and accurate as well as nice repetition in the determination of AFB1 content in edible oils.更多还原
