纳米TiO2对人淋巴细胞DNA损伤的研究

【摘要】 目的探讨一维及三维纳米TiO2和常规TiO2的遗传毒性。方法人离体淋巴细胞培养,采用不同剂量三种不同材料的TiO2染毒,通过观察淋巴细胞转化率,并结合单细胞凝胶电泳实验、微核实验测定细胞DNA损伤。结果淋巴细胞转化率各染毒组与阴性对照差异为无统计学意义;单细胞凝胶电泳实验显示1D TiO2拖尾率与常规TiO2相比差异无统计学意义,3D TiO2与常规TiO2之间的差异有统计学意义(P<0.05);TiO2染毒各个剂量组的微核率与阴性对照差异均无统计学意义。结论3D TiO2具有一定的遗传毒性,而1D TiO2表现不明显。

【Abstract】 Objective To explore the genetic toxicity of one-dimensional and threedimensional nano-TiO2.Methods Human lymphocytes were cultured and exposed to different doses of TiO2.DNA damage was observed by lymphocyte transformation rate determination,comet assay,and micronucleus test.Results There was no significant difference in lymphocyte transformation rate compared with the negative controls;the difference of tailing rate was only seen between 3D TiO2 and conventional TiO2(P<0.05)in comet test;and there was also no difference in micronucleus rate among different groups.Conclusions The results shows that only 3D TiO2 has some genetic toxicity,while the 1D TiO2 seems no those effects.
