survivin基因启动子在多种肿瘤细胞株中的转录活性研究
Transcriptional Activity of Survivin Promoter in Tumor Cell Lines
【摘要】 目的:研究两个长度不同的survivin基因启动子在多种肿瘤细胞株中的活性。方法:利用脂质体将不同长度的sur-vivin基因启动子调控荧光素酶的报告基因载体瞬时转染HepG2、Lovo、SW480、AGS肿瘤细胞和3T3正常细胞,48 h后经荧光素酶检测,比较两个survivin基因启动子的活性。结果:两个不同长度的survivin基因近端启动子在多种肿瘤细胞株中均显示出较高的活性,其中以pGL3-Surp340的活性最高。结论:实验中使用的两个survivin启动子片段肿瘤细胞中有转录活性,在3T3正常细胞中活性低。其中survivin340启动子的活性明显高于survivin1430启动子,其有可能作为调控元件用于肿瘤的靶向性基因治疗。
【Abstract】 Objective:To study the transcriptional activity of two different lengths survivin gene promoter in a variety of tumor cell lines.Methods: The pGL3-Surp340 and pGL3-Surp1430 plasmid were transiently transfected into human tumor cells: HepG2,Lovo,SW480,AGSand normal cell lines 3T3.The transcriptional activities of Survivin promoter in various cells were determined by measuring the luciferase activities after 48hours of transfection.The pGL3-CMVp(containing the CMV promoter) was used as positive control.Result: Luciferase assay showed that the transcriptional activities of the two survivin promoters in tumor cells were extremely higher than normal cells,and the transcriptional activity of pGL3-Surp340 was higher than pGL3-Surp1430.Conclusion: The two survivin promoter is more highly activated in tumor cells than in normal cells,which may be a potential candidate promoter in the gene therapy of tumor.
