胡桃醌对人肝癌BEL-7402细胞增殖和膜流动性的影响

【摘要】 目的:研究胡桃醌诱导人肝癌BEL-7402细胞凋亡及其作用机制。方法:采用MTT法测定细胞生长抑制率,通过流式细胞仪观察凋亡细胞,采用荧光偏振技术离体动态地检测30 min内胡桃醌对BEL-7402细胞各向异性r、荧光偏振度P和微粘度η的影响,分析细胞膜流动性的改变。结果:各浓度的胡桃醌均可抑制人肝癌BEL-7402细胞的生长,诱导其凋亡,IC50为13.78μM。不同浓度胡桃醌均可引起BEL-7402细胞膜的r、P和η值显著性降低(P<0.05),尤其在前1 min内r、P、η值下降迅速,1min后各指标下降的趋势渐渐平缓。结论:胡桃醌能显著增加BEL-7402细胞的膜流动性,促进其本身进入细胞内部,抑制细胞的增殖,诱导细胞凋亡。

【Abstract】 Objective:To study the effect of juglone on inducing apoptosis of human liver cancer BEL-7402 and its mechanism.Methods: MTT method was used to determine cell inhibiting rate,and flow cytometry was used to observe cell apoptosis.The fluorescence depolarization method was used to measure values of fluorescence anisotropy,fluorescence polarization as well as microviscosity in BEL-7402 cells and microviscosity in liposome continuously for 30 min.Results: The results showed that juglone at diferent dosages could inhibit the growth of human liver cancer BEL-7402,and induce cell apoptosis,IC50 were 13.78μM.Juglone induced a significant decrease of fluorescence anisotropy,fluorescence polarization as well as microviscosity in BEL-7402 cells,particularly in the first 1 min.After 1 min,the values of anisotropy were remained lower levels.Conclusion: Juglone could induce apoptosis of human liver cancer BEL-7402 and significantly increase membrane fluidity in BEL-7402 cells and sequentially make it easier that juglone came into the cell to induce tumor cell apoptosis.
