姜黄素对人子宫内膜癌细胞增殖和凋亡的影响

【摘要】 目的:探讨姜黄素对人子宫内膜癌细胞(HEC-1B)增殖和凋亡的影响。方法:应用6.67~66.67μmol/L的姜黄素分别处理HEC-1B 48h后,采用MTT法检测姜黄素对HEC-1B的增殖程度的影响;流式细胞仪进行细胞周期时相分析;荧光显微镜观察细胞核形态的变化。结果:①MTT法显示培养48h后,姜黄素组的吸光度值A490nm低于对照组(P<0.01),且在一定浓度范围内呈剂量依赖性;②流式细胞仪检测显示培养48h后,姜黄素组的G2/M比例高于对照组(P<0.05),且在一定浓度范围内呈剂量依赖性;③荧光显微镜下给药组部分细胞发生凋亡形态学改变,凋亡率为10.22%~34.72%。结论:姜黄素可抑制HEC-1B的增殖,诱导细胞凋亡,其诱导凋亡的机制可能是通过将细胞阻滞在G2/M期来实现的。

【Abstract】 Objective:To investigate the effect of curcumin on the proliferation and apoptosis of endometrial carcinoma cell line HEC-1B.Methods: After HEC-1B cells were treated with 6.67~66.67μmol/L curcumin for 48h,HEC-1B proliferation was determined with MTT colorimetric method;The cell cycle distribution was detected by flow cytometic analysis;By using fluorescence microscope,the morphologic alterations of HEC-1B nucleus were observed.Results: ①MTT results showed that,the values of A490 nm in curcumin groups were lower than that in the control group at hours 48(P<0.01),and the effect for HEC-1B proliferation gradually decreased with the increasing content of curcumin;②Flow cytometry results showed that,the values of G2/M in curcumin groups were higher than that in the control group at hours 48(P<0.01),and the effect for cell cycle gradually increased with the increasing content of curcumin;③Partial cells presented the characteristic morphological changes of apoptosis under the fluorescent microscope;the apoptosis rates were 21.5%~33.5%.Conclusion: Curcumin could significantly inhibit the growth and proliferation of HEC-1B lines in vitro.Apoptosis induced by curcumin was the major mechanisms of curcumin.The influence of the distribution of cell cycle phase was associated with apoptosis.
