鹿茸多肽对兔骨性关节炎软骨细胞增殖及凋亡调节作用的实验研究

【摘要】 目的观察鹿茸多肽对骨性关节炎细胞增殖和凋亡的保护、调节作用。方法建立兔骨性关节炎模型,体外分离培养软骨细胞,以假手术组细胞为正常对照组细胞,造模组细胞为骨关节炎软骨,分别加入6.25μg/mL鹿茸多肽为低剂量组,加入12.5μg/mL鹿茸多肽为中剂量组,加入25μg/mL鹿茸多肽为高剂量组,通过流式细胞仪检测骨性关节炎细胞增殖及凋亡。结果与正常对照组比较,模型组G0/G1期细胞比例显著降低(P<0.01),G2/M和S期细胞比例明显升高(P<0.05)。鹿茸多肽对骨关节炎软骨细胞周期各时相并无明显的调控作用,可明显降低早期凋亡细胞比例,且有一定的量效关系。结论鹿茸多肽对骨性关节炎软骨细胞的早期凋亡有明显抑制作用。

【Abstract】 Objective To observe the protection and regulation of velvet antler polypeptides(VAP)on cell proliferation and apoptosis in osteoarthritis(OA).Methods OA models were established in rabbits.Chondrocytes were isolated and cultured in vitro.The cells in sham-operation group were taken as the cells in normal control groups.OA chondrocytes were the cells in model group.Low-dose group,middle-dose group and high-dose group were designed by adding VAP 6.25 μg/mL,12.5 μg/mL and 25 μg/mL successively.The flow cytometry was used to determine the proliferation and apoptosis of OA cell.Results Compared with normal group,the cell proportion at G0/G1 phase in model group decreased significantly(P<0.01),that at G2/M and S phases increased apparently(P<0.05).VAP had no apparent regulation and control on OS chondrocyte circle at each phase.VAP reduced apparently the proportion of cell apoptosis at early phase and presented a certain dose-effect relationship.Conclusion VAP inhibits significantly the early apoptosis of chondrocytes in OA and presents the protection of chondrocytes.
