含RGD多肽的重组丝蛋白的设计与合成

【摘要】 利用基因工程方法将含短肽RGD的氨基酸序列连接到桑蚕丝素蛋白中,设计并合成了具有[TS(GAGAGS)5GAGYT-GRGDSPAGYGAGVGAS]n一级结构的重组桑蚕丝素-RGD融合蛋白(简称Silk-RGD),利用SDS-PAGE、红外光谱、质谱和热重分析等手段对所得产物进行了分析。结果表明:重组融合蛋白与所设计的氨基酸序列一致,并且体现出了天然桑蚕丝蛋白的结构与性能。由于重组蛋白含RGD序列预期具有良好的细胞相容性,因此该融合蛋白材料有望在生物医用材料、组织工程等领域得到应用。

【Abstract】 A novel hybrid protein(Silk-RGD)which combines the RGD tripeptide and primary amino acid sequence derived from native Bombyx mori silk fibroin was designed and synthesized by using the transgenic Escherichia coli expression system.The obtained protein was characterized by using SDS-PAGE,FTIR,mass spectrum and TG analyses.The data showed that the amino acid sequence of the recombinant hybrid protein agreed well with the designed one,and the protein showed the similar structure and properties with the native silk fibroin.Since it contains the RGD sequence,this hybrid protein is expected to be cell biocompatibility,which may be an ideal candidate for the future application in biomaterial and tissue engineering fields.
