益气化瘀补肾方对大鼠气虚型颈椎病的作用机制研究

【摘要】 目的:探讨益气化瘀补肾方对气虚型颈椎病大鼠的作用机制。方法:选择3月龄雌性SPF级SD大鼠30只,随机分为正常组、气虚型颈椎病模型组(模型组)和益气化瘀补肾方组(治疗组),每组10只。正常组不施加任何手术,气虚型颈椎病模型采用动静力失衡性大鼠颈椎间盘退变模型复合疲劳加饥饱失常法气虚模型建立,手术3个月后治疗组大鼠予益气化瘀补肾方灌胃治疗1个月。通过放射免疫学、颈椎间盘组织病理学、免疫组织化学、分子生物学等技术研究益气化瘀补肾方作用机制。结果:模型组动物出现精神委靡、少动、体质量下降等气虚表现,cGMP降低,cAMP/cGMP增高,三酰甘油和CD62p增高,E2降低,颈椎间盘退变,Ⅱ型胶原减少,Agc1、Col2a1表达减少,MMP-13表达增高;治疗组与模型组相比,动物精神状态恢复稍快,cAMP/cGMP增高,三酰甘油和CD62p降低,颈椎间盘退变有所改善,Ⅱ型胶原增加,Col2a1和TIMP-1表达增高。结论:益气化瘀补肾方通过免疫、代谢、凝血等系统改善证候指标并延缓椎间盘退变。

【Abstract】 Objective:To explore the mechanism of "Yiqi Huayu Bushen Recipe" (YHBR) ,a compound traditional Chinese herbal medicine,in treating cervical spondylopathy (CS) of qi deficiency pattern in rats. Methods:A total of 30 three-month-old female Sprague-Dawley rats were randomly divided into normal control,untreated group and treated group;there were ten rats in each group. The rat model of cervical spondylopathy of qi deficiency pattern was established by combining disease and pattern models. After one-month YHBR treatment in treated group,all rats were sacrificed. Serum,plasma and cervical intervertebral discs were detected and observed by radio-immunology,histopathology,immunohistochemistry,etc. Results: Compared with those in the normal control group,rats in the untreated group displayed symptoms due to deficiency of vital energy such as tiredness,ptosis,few movement,body weight decreased,cGMP decreased and cAMP/cGMP increased,the content of triglyeride and expression of CD62p were increased,E2 decreased and intervertebral disc structure destructed,typeⅡcollagen protein expression decreased,the expressions of Agc1 and Col2a1 mRNAs decreased and that of MMP-13 increased. Compared with the untreated group,rats in treated group status recovered more quickly,triglyeride and CD62p decreased,intervertebral disc structure was improved,the expressions of type Ⅱ collagen protein in annulus fibrosus and Col2a1 and TIMP-1 mRNAs also increased. Conclusion: YHBR may alleviate symptoms and delay degeneration of cervical intervertebral disc from immune system,metabolism system and coagulation system to treat CS of qi deficiency pattern.
