一种检测粪便隐血的改良化学法

【摘要】 目的建立一种检测粪便隐血的改良化学法。方法用3,3,′5,5′-四甲基联苯胺(TMB)为主要原料制备成干片试纸,将试纸浸入粪便样本液,试纸呈蓝色即为阳性。结果能够检测到样本中血红蛋白(Hb)含量范围为5~10 000μg/mL,消化道中蛋白酶对Hb的消化对本法无影响。高浓度Hb样本能够引起免疫法钩状效应,在本法中无影响。结论改良的TMB方法要比目前常规使用的化学法更有优势,快速、简便、能够单人份检测,尤其是检测的灵敏度高和检测范围宽。在免疫法粪便隐血试验中同时应用化学法可以检出假阴性反应。

【Abstract】 ObjectiveTo establish an improved chemical method for the determination of fecal occult blood test(FOBT).MethodsA dry FOBT reagent was prepared using 3,3′,5,5′-tetramethylbenzidine(TMB) as main matieral.ResultsThe range of detection of hemoglobin(Hb) was 5-10 000 μg/mL.Digestion of Hb by enzymes in gastrointestinal(GI) tract did not effect the FOBT detection with improved method.High concentration of Hb,which showed negative result immunological FOBT by hook effect,and had no influence in chemical FOBT.ConclusionsChemical methods are need to be used in FOBT.The improved TMB method is better than traditional chamical methods.It is rapid,simple and increases the sensitivity and the detection range.In FOBT chemical method should be used in addition of immunological method for avoiding false negative result.
