【摘要】 目的探讨基质金属蛋白酶-2(MMP-2)和组织基质金属蛋白酶抑制剂-2(TIMP-2)在人胃癌组织中的表达,及其与胃癌临床病理特征的关系。方法选取100个胃癌手术切除标本中胃癌组织、癌旁组织及正常胃组织,采用免疫组织化学法检测MMP-2和TIMP-2蛋白的表达,并分析两者的表达及其与胃癌临床病理特征之间的相关性。结果胃癌组织中的MMP-2蛋白阳性率为77.0%,显著高于癌旁组织的33.0%和正常胃组织的8.0%(P值均<0.01),癌旁组织又显著高于正常胃组织(P<0.01)。胃癌组织中的TIMP-2蛋白阳性率为29.0%,显著低于癌旁组织的62.0%和正常胃组织的76.0%(P值均<0.01),癌旁组织又显著低于正常胃组织(P<0.01)。随着肿瘤浸润深度的增加(Tis～T4)、周围淋巴结转移的增多(N0～N3)、临床分期的进展(0～Ⅳ期),MMP-2蛋白阳性率逐渐升高,TIMP-2蛋白阳性率逐渐降低,且差异有统计学意义(P值均<0.01)。结论MMP-2和TIMP-2的表达可能与胃癌的发生、侵袭和转移有关,检测MMP-2和TIMP-2的表达有助于预测胃癌转移。更多还原

【Abstract】 Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in human gastric cancer and its relationship with the clinicopathological properties of human gastric cancer. Methods A total of 100 surgical specimens were investigated by S-P immunohistochemical staining using an anti-MMP-2 monoclonal antibody and an anti-TIMP-2 polyclonal antibody. The expression of MMP-2 and TIMP-2 in various regions of the gastric cancer tissues,paracancerous tissues and normal tissues was examined,and its relationship with clinicopathologic properties were analyzed. Results The expression of MMP-2 in the gastric cancer tissues was significantly higher than that in the paracancerous tissues and normal gastric tissues (P<0.01); that in the paracncerous tissues was significantly higher than that in the normal tissues (P<0.01). TIMP-2 expression in the gastric tissues was significantly lower than that in the paracancerous tissues and normal gastric tissues(P<0.01); that in the paracncerous tissues was significantly lower than that in the normal tissues (P<0.01). The expression of MMP-2 increased and the expression of TIMP-2 decreased with the invasion depth of tumors (Tis—T4),the lymph node metastasis (N0—N3),and the progression of clinical staging (0—Ⅳ,P<0.01). Conclusion MMP-2 and TIMP-2 expresion may play an important role in the carcinogenesis,invasion and metastasis of gastric carcinoma. Examination of MMP-2 and TIMP-2 expression may help the predication of gastric cancer metastasis.更多还原
