【摘要】 目的从山楂中提取及鉴定谷甾醇,并研究其对3种肿瘤细胞和人体正常肝脏细胞的作用。方法取对数生长期的细胞进行MTT法细胞增殖实验及DNA凝胶电泳实验,观察谷甾醇对不同细胞的作用。结果MTT法细胞增殖实验显示一定剂量的谷甾醇对体外培养的HepS、S180、EAC细胞有显著的抑制作用(P<0.01),抑制率总体上与时间和浓度呈正相关;对L02细胞无明显的抑制作用(P>0.05);电泳结果显示经谷甾醇作用的HepS细胞DNA出现梯带。结论谷甾醇可能是通过促进肿瘤细胞凋亡来抑制其增殖。更多还原

【Abstract】 Purpose To extract and identify sitosterol from hawthorn fruits to study its anticancer effect on several cells.Methods Designing experiments of cells proliferation and DNA agarose gel electrophoresis.Results The proliferation experiments showed that sitosterol has an inhibiting effect on HepS,S180,EAC cell(P<0.01),except L02 cells.And the inhibitory ratehad a direct ratio to the value of time and concentration.DNA ladders were found in DNA agarose gel electrophoresis results with HepS cells.Conclusion Sitosterol can inhibit proliferation of cancer cells,probably by causing apoptosis.更多还原
