腹腔镜气腹环境下前列腺癌细胞HIF-1α,VEGF和Cadherin的表达变化

【摘要】 目的研究模拟腹腔镜气腹环境对前列腺癌(PC-3)细胞的影响,探讨可能的分子机制及影响的时效。方法PC-3细胞随机分为3组:常氧培养组、99%CO2培养组、氯化钴(CoCl2)预处理后常氧培养组。采用Westernblot检测3组的HIF-1α,VEGF,Cadherin变化情况。结果99%CO2培养2、4、8h后,可检测到HIF-1α,VEGF的表达逐步上升,Cadherin逐步下降,与常氧培养组有显著差异(P<0.05),组内统计分析三指标变化具有相关性。氯化钴预处理组观察到相似变化。将99%CO2培养4h后的细胞恢复常氧培养至16h,所见改变基本恢复。结论CO2模拟气腹环境可通过缺氧导致HIF-1α高表达,从而引起肿瘤转移相关因子VEGF,Cadherin蛋白表达水平升高。气腹作用4h内所引起上述因子改变是可逆的。

【Abstract】 Objective To investigate the influence of CO2-insufflation on the prostate cancer cells (PC-3) and explore its molecular mechanism and influencing of time-related effect. Methods With an artificial pneumoperitoneum model in vitro,prostate carcinoma cells were divided into 3 groups:conventional culture group (A),99% CO2-insufflation culture group (B) and cobalt chloride-preprocessing conventional culture group (C).Expression of HIF-1α,VEGF and Cadherin were measured at 2,4,and 8 hours by Western blot. Results The expression of HIF-1α,VEGF and Cadherin in prostate carcinoma cells changed significantly. Changed with time,the expression of the HIF-1α and VEGF increased gradually in group B,while Cadherin decreased,compared with group A (P<0.05).Statistical analysis showed relationship among the 3 parameters in group A. There were similar changes in group C.No significant changes were observed in prostate carcinoma cells with conventional culture for 16 h,which were pre-cultured in group B for 4 h. Conclusions The high expression of HIF induced by CO2-insufflation may affect genetic transcription and protein expression related to tumor metastasis. The influence of CO2-insufflation within 4h is reversible in vitro.
