利用RAPD标记分析百合种质的遗传多样性
Genetic Diversity Analysis of Lilium spp. Germplasms by RAPD
【摘要】 中国是百合属植物的起源中心,栽培和应用历史悠久。如何评价及充分利用我国丰富的百合资源是选育优质百合品种的一个重要课题。本研究以全国各地收集获得的45份百合种质(包括21份野生种和24个栽培品种)为材料,利用RAPD技术对这些百合材料进行遗传多样性的分析及亲缘关系的鉴定。结果表明:从300条引物中筛选获得29条可重复多态性引物(1条野生种特异引物:S249引物)。以45份百合材料总DNA为模板29条引物共扩增出398个多态性位点。UPGMA聚类分析表明:45份材料的遗传距离在0.158~0.504,平均遗传距离是0.370。45份材料可以分为5类:包括云南地区野生种、铁炮百合杂交系、亚洲百合杂交系、东方百合杂交系和其他地区野生种。
【Abstract】 China has a long history of lilies’cultivation and application as the lily origin center. How to evaluate and make use of abundant lily germplasms are important for the breeding commercial lily cultivars with better agronomic characteristics. In this study, the genetic diversity of 45 lily germplasms (21 wild species and 24 cultivars) was analyzed by the RAPD markers. Twenty eight polymorphic primers and one primer specific to wild species were selected from 300 random primers, which generated 398 polymorphic bands in total. The dendrogram tree obtained by UPGMA method (Unweighted Pair-Group Method of Arithmetic Average) showed that the range of the genetic distances among all the germplasms were between 0.158 and 0.504, and the average is 0.370. All of the 45 lily accessions were clustered into 5 groups: the wild species from Yunnan, the Longiflorum hybrids, the Asian hybrids, the oriental hybrids and the wild species from other provinces in China.
