茜素红二级散射法测定核酸

【摘要】 探讨在阳离子表面活性剂十六烷基三甲基溴化铵(CTMAB)存在下茜素红(AR)二级散射法测定脱氧核糖核酸(DNA)的方法,研究在pH=2.8的B-R缓冲溶液中AR-CTMAB-DNA体系的光谱特征、影响因素和最佳反应条件。在最佳条件下二级散强度与DNA线性范围为0~2.2 mg.L-1,检出限为1.12×10-6g.L-1,用该法测定合成样品DNA的结果,相对标准偏差0.70%,回收率为101.5%。

【Abstract】 It is significant for the quantitative determination of deoxyribonucleic acid(DNA) in biology and clinical medicine.An introduction on the quantitative analysis of DNA with alizarin red(AR) is presented in detail,especially pertaining to the determination of DNA and its applications using second-order scattering method under cation surfactant cetyltrimethylammonium bromide(CTMAB) and pH=2.8 B-R buffer solution.Spectrum characteristic,effect factors and optimum reaction conditions of AR-CTMAB-DNA system were studied.The linear range is 0~2.2 mg·L-1;the detection limit is 1.12×10-6 g·L-1.This method was applied to determine DNA in synthesized samples with simplicity and rapidity.The relative standard deviation was 0.70 %,and the recovery was 101.5 %.
