【摘要】 目的:研究小鼠胸膜间皮细胞免疫相关基因表达谱的状况,从而探索用现代免疫及基因芯片技术评价解释中医肺气学说的新方法。方法:取8只BALB/c小鼠胸膜组织块剪切成碎块,提取mRNA,经标记、杂交、清洗、芯片扫描、图像采集与数据分析等过程获得基因表达谱。结果:共筛选出小鼠胸膜间皮细胞53条免疫相关高表达基因。其中主要有免疫应答高表达基因14条;炎性反应高表达基因6条;非特异性免疫应答高表达基因7条;免疫应答调节高表达基因4条;急性反应阶段高表达基因3条;抗原加工与提呈高表达基因2条;Ⅱ型超敏反应正向调节高表达基因2条。结论:从现代免疫学观点及正常小鼠胸膜间皮细胞基因芯片的研究来看,胸膜间皮细胞既表达免疫相关的正向调节基因,亦表达负向免疫相关的调节基因。这不仅可为现代免疫学增添新的内容,而且对丰富中医肺气学说,研究中医肺脏的实质乃至推动中医学理论的深入研究都将具有积极意义,为胸膜的其他后续研究构建一平台,为丰富中医肺气学说与肺气的实质提供分子生物学的基础研究依据;弥补这方面的研究空白。更多还原

【Abstract】 Objective:To investigate condition of mice pleural mesothelial cell(PMC)immunity-related gene expression spectra,accordingly to explore modern immunity and gene chip technique to evaluate Traditional Chinese Medicine Lung-Qi theory with the latest method.Methods:Using DNA chip to detect the mRNA form 8 mice tissues of pleural mesothelial cell(PMC).Results:53 immunity-related hypso-genes expression were picked out in mice PMC,including 14 immune response and immune regulatory hypso-express genes,6 inflammatory reaction hypso-express genes of immune response,3 acute reaction period hypso-express genes,2 antigen processing and presentationhypso-express genes,2 up-regulation hypso-express genes of Ⅱ type hypersensitivity reaction.Conclusions:PMC not only expressimmune-associated up-regulation hypso-express genes,but also hypso-express immune-associated down-regulation hypso-expressgenes.To hint pleura may be one immune coordinator,there will be molecule biological underlaying research identification to be provided for an abundance Lung-Qi parenchyma of Traditional Chinese Medicine,as well as there will be new research thinking to be provided.更多还原
