针刺对胰岛素抵抗模型大鼠肝脏脂毒性损伤的影响

【摘要】 目的:观察针刺对模型大鼠胰岛素抵抗(IR)状态的调整作用,及肝脏游离脂肪酸(FFA)含量和形态学的影响。方法:将24只大鼠随机分成空白组、模型组、针刺组各8只。空白组用普通饲料喂养,其余两组采用高脂高糖高盐饲料喂养复制IR模型成功后,针刺组给予针刺治疗,1次/天,共2周。治疗结束后检测FBG、INS、FFA、计算ISI,并做肝脏病理切片。结果:①与空白组比较,模型组大鼠FBG、INS显著升高,ISI显著降低(均P<0.01);与模型组比较,针刺组大鼠FBG、INS下降,ISI上升(均P<0.01);②与空白组比较,模型组大鼠肝脏FFA含量显著升高(P<0.01);与模型组比较,针刺组大鼠脏FFA含量显著降低(P<0.01),与空白组比较无显著性差异;③病理结果显示:模型组肝细胞存在明显的脂肪变性和水样变性,针刺组则程度较轻。结论:针刺可以改善模型大鼠的IR状态,并减轻其肝脏脂毒性损伤,其作用途径之一可能是通过减轻IR大鼠肝脏FFA的生成和堆积来实现的。

【Abstract】 Objective: To observe the effect of acupuncture on insulin resistance(IR) state,the content of free fatty acid(FFA) and hepatic morphological changes.Methods: A total of 24 rats were randomly divided into blank group,model group and acupuncture group.Each group had 8 rats.The insulin resistance models were induced by feeding high-fat-sugar-salt forage.After the models were successfully established,rats in acupuncture group accepted acupuncture of two weeks,once a day.The fasting blood glucose(FBG),plasma insulin(INS),insulin sensitivity index(ISI),FFA content and morphological changes of the liver in each group were observed.Results: 1.The levels of FBG and INS in model group increased markedly and ISI levels decreased significantly as compared with blank group (P<0.01);The levels of FBG and INS in acupuncture group decreased and ISI levels increased as compared with model group(P<0.01).2.FFA content in model group increased as compared with blank group(P<0.01),and decreased significantly in acupuncture group as compared with model group(P<0.01).3.There were significant adipose and hydropic degeneration on hepatic cells in model group,but this change was not obvious in acupuncture group.Conclusion: Acupuncture can improve the IR state of modelrats and reduce the fatty-toxicity-damage.One of the mechanisms may be related with the reduction of FFA production and accumulation.
