从发酵液中直接提取柚苷酶

【摘要】 用双水相萃取方法直接从发酵液中提取柚苷酶进行了实验,就聚乙二醇-硫酸铵双水相系统中的聚乙二醇相对分子量、聚乙二醇浓度、硫酸铵浓度、无机盐的添加等因素对柚苷酶的分配行为的影响进行了探索。结果显示:在用浓度为14%~16%的PEG1000与浓度为14%~18%硫酸铵和0.1%氯化钠组成的双水相体系中,柚苷酶的分配系数达1.85以上,酶活力回收率达80%,提纯倍数为2.1。

【Abstract】 The naringinase was extracted by aqueous two-phase from fermentation liquor.The effect produced by the distributing of naringinase that was caused by such factors as the relative molecular weight of PEG,the consistency of PEG,the consistency of ammonium sulfate and the appending of mineral salt in the aqueous two-phase system of PEG-ammonium sulfate was explored.The result showed that the coefficient of naringinase was more than 1.85,the recovery rate of enzyme activity was up to 80% and the purifying multiple was 2.1 in the aqueous two-phase system that was composed of PEG 1 000 of 14%~16%,(NH4)2SO4 of 14%~18% and NaCl of 0.1%.
