HCC细胞膜相关蛋白的快速鉴定

【摘要】 为快捷、特异地鉴定HCC(hepatocellular carcinoma,人肝细胞癌)细胞的膜相关蛋白,用DOC(脱氧胆酸钠)对贴壁培养的HCC细胞系HepG2进行处理,使细胞皱缩并脱离培养面,收集未脱离的HepG2膜相关蛋白,经过聚丙烯酰胺凝胶电泳分离,最后应用ESIQUAD-TOF MS(electrospray ionization quadrupole time-of-flight mass spectrometry),成功鉴定出包括热休克蛋白90α、玻璃体粘连蛋白前体、层粘连蛋白α5亚单位前体、胶原蛋白前体、鳞状细胞癌T细胞识别抗原-3等35个蛋白质,其中膜相关蛋白18个,所占比例超过50%。

【Abstract】 In order to swiftly and specially identify the membrane-associated proteins of HCC (hepatocellular carcinoma) cells, DOC (sodium deoxycholate) has been employed to shrink HepG2 cells and to detach the cells from the surface of the flask on which the membrane-associated proteins remain. The membrane-associated proteins, after collected, were seperated by polyacrylamide gel electrophoresis and then were identified by ESI-QUAD-TOF MS (electrospray ionization quadrupole time-of-flight mass spectrometry). Finally, 35 proteins, including Heat shock protein HSP 90-alpha, Vitronectin precusor, Laminin subunit alpha-5 precursor, Collagen alpha-6(Ⅳ) chain precursor and Squamous cell carcinoma antigen recognized by T-cells 3, were successfully identified and eighteen membrane-associated proteins ocuupied more than 50% of the identified proteins.
