油菜抗菌肽的分离及抑菌机制的初步研究
Isolatoin of the antifungal peptide from oil-rape(Brassica napus)seed and study on antifungal mechanism
【摘要】 通过Sephadex G-100凝胶柱层析和HPLC的方法,从油菜籽的水溶性物质中初步分离一种活性肽。采用抑菌圈法测定活性肽的抑菌活性,结果表明,这种活性肽对病原性真菌有明显的抑制作用,对细菌没有抑制效果。对活性肽的稳定性测定表明,活性肽加热到90℃未丧失活性,在pH7的时候抑菌效果最为明显。利用扫描电镜观察到这种活性肽能够很明显影响真菌孢子正常生理形态。
【Abstract】 An active peptide was isolated from the soluble exacts of oil-rape seeds with Sephadex G-100 gel chromatography column and HPLC. Through fungicidal activity bioassay, the peptide inhibited the growths of epiphyte but not bacteria. Through researches of stability of the peptide to heat, acid and alkali, the peptide had an activity to epiphyte up to 90 ℃, and inhibited epiphyte most strongly at pH 7. By scanning electronic microscope (SEM), it was observed that the active peptide changed epiphyte spore morphology obviously.
