SD乳鼠心肌细胞的培养及其低温下钙波-搏动耦联的初步研究
Culture of the cardiac myocytes and preliminary study on coupling of calcium wave and cardiac myocytes beating at low temperature
【摘要】 分离培养3d以内SD乳鼠心肌细胞,Fluo-4负载后分别于37℃,24℃环境中在激光扫描共焦显微镜(Laser Scanning Confocal Microscopy,LSCM)下进行细胞搏动及胞内Ca2+运动的观测。结果显示,心肌细胞在体外呈集落生长。当37℃时,集落中无钙波产生,但集落细胞同步搏动;当24℃时,集落中个别细胞先发生钙波,随后集落细胞才同步搏动。证明心肌细胞搏动与胞内Ca2+浓度变化相关,在24℃低温下个别细胞产生的钙波可能对其所在集落细胞的同步搏动有诱导并维持其搏动的作用。
【Abstract】 Isolated and cultured the cardiac myocytes from SD neonatal rat (<3 days). After loaded with Fluo-4 fluorescence probes, Ca2+ motion of cardiac myocytes was observed by laser scanning confoeal microscope (LSCM) in 37 ℃ and 24 ℃ respectively. It was observed that the in vitro cultured cardiac myocytes formed colony. In 37 ℃, cardiac myocytes in the same colony were in synchronous pulsation. In contrast, in 24 ℃, calcium wave first appeared in some cardiac myocytes of the samecolony, then cardiac myocytes in the same colony were beating synchronously. The results showed that the beating of cardiac myocytes was related to the change of Ca2+ concentration. Furthermore, in the relative low temperature (24 ℃), calcium wave happened in some cardiac myocytes may induce and maintain the synchronous beating of cardiac myocytes in the same colony.
