Lrrc10在体外心肌发生过程中的表达研究
Expression of Lrrc10 during Cardiogenesis in vitro
【摘要】 Lrrc10是一心脏特异新基因。为探讨Lrrc10在心肌细胞分化过程中的表达,采用DMSO诱导P19细胞分化,建立体外心肌发生模型,显微镜观察细胞形态变化,以心α-MHC和β-MHC作为心肌分化的标志,RT-PCR检测分化过程中各基因的表达。结果表明,P19细胞经诱导后形态发生明显变化;α-MHC在诱导分化6d时开始表达,β-MHC在诱导分化8d时开始表达,表明心肌分化成功。而Lrrc10在诱导分化2d时就开始表达,提示其功能可能与心肌分化有关。
【Abstract】 Lrrc10 is a novel heart specific gene.In order to investigate the expression of Lrrc10 during cardiogenesis,an in vitro cardiogenesis model was developed by using P19 cells which underwent differentiation induced by DMSO.Morphological change of cells was observed by microscope,α-MHC and β-MHC,used as the markers of cardiogenesis expression of genes during the differentiation,were determined by RT-PCR.The results showed that P19 cells exhibited significant morphological changes after induced by DMSO.Expression of α-MHC was detected after differentiated for 6 days and β-MHC was detected after differentiated for 8 days.These results suggested that differentiation of P19 cells in vitro succeeded.Moreover,expression of Lrrc10 was detected by RT-PCR at the second day of differentiation and this suggested that Lrrc10 probably plays a role in cardiogenesis.
