龙血竭对组织工程皮肤修复皮肤缺损的作用
Effect of Resina Draconis on the Repair of Skin Defect Treated with Tissue Engineering Skin
【摘要】 【目的】观察龙血竭对组织工程皮肤修复皮肤缺损创面的促进作用。【方法】将人成纤维细胞种植入胶原海绵,人角质形成细胞种植在表层,构建组织工程皮肤,并移植入裸鼠皮肤缺损创面;实验动物随机分为4组,选用"活血圣药"龙血竭于移植后2d分别进行创面外敷、单纯口服、创面外敷+口服治疗,并设空白对照组。组织工程皮肤移植后9d取材检测,观察创面局部表皮厚度、真皮层中层粘连蛋白(Ln)、Ⅰ型胶原蛋白的表达及毛细血管增生情况。【结果】龙血竭能促进组织工程皮肤移植后表皮的发育及真皮层中毛细血管的增生,增强Ln、Ⅰ型胶原蛋白的分泌,龙血竭外敷+口服组效果最佳。【结论】龙血竭对组织工程皮肤修复皮肤创面有促进作用。
【Abstract】 Objective To investigate the effect of Resina Draconis(RD)on the repair of skin defect treated with tissue engineering skin.Methods The tissue engineering skin was constructed with collagen sponge as the dermal scaffolds where human skin fibroblasts were transplanted inside and human keratinocytes were transplanted on the surface.Then the tissue engineering skin was transplanted into the skin wounds in naked mice with skin defects.After transplatation for 2 days,RD was used for the treatment.According to the administration method of RD,the treated mice were randomized into 3 groups:external-application RD group,oral-use RD group and combination group(external application and oral use of RD).A blank control group receiving tissue engineering skin transplantation alone was also set up.The differentiation tissue of the tissue engineering skin was sampled after transplantation for 9 days.And epidermal thickness of focal wound skin,laminin(Ln)and collagen typeⅠprotein expression in the dermis and the growth of the capillaries were observed.Results Resina Draconis promoted the development of epidermis and the growth of capillaries in dermis after transplantation of the tissue engineering skin,and increased the protein expression of Ln and collagen typeⅠ,the above effect being the best in the combination group.Conclusion Resina Draconis shows an enhancement on the repair of skin defect treated with tissue engineering skin.
