广藿香愈伤组织诱导和分化再生植株的研究
Studies on Callus Induction and Plantlets Regeneration of Pogostemon cablin(Blanco)Benth.
【摘要】 【目的】开展广藿香愈伤组织的诱导及分化再生植株的研究,建立高效的组织培养再生系统。【方法】以广藿香叶片、带节茎、不带节茎及根尖为材料进行离体培养,筛选影响愈伤组织诱导、增殖及分化的因素。【结果】6-苄氨基嘌呤(BA)或N-苯基-N-1,2,3-噻二唑-5-脲(TDZ)与α-萘乙酸(NAA)配合有利于愈伤组织的诱导,BA的适宜浓度为0.2~0.5 mg/L;4种外植体中,带节茎及不带节茎较易诱导出愈伤组织。BA和NAA配合使用能较好地促进愈伤组织的增殖;培养30 d的愈伤组织较60 d的具有更强的增殖和分化能力。BA有利于愈伤组织的分化;叶片诱导的愈伤组织分化能力较强。【结论】建立了高效的广藿香愈伤组织诱导及分化再生植株的离体再生体系。
【Abstract】 Objective Callus induction and plantlets regeneration of Pogostemon cablin(Blanco)Benth.are studied to establish a high-efficiency regeneration system for tissue culture.Methods The leaf segments,nodular stem segments,stem segments and root tips from Pogostemon cablin(Blanco)Benth.were cultured in vitro.The factors which affect the induction,multiplication and differentiation of callus were studied.Results Benzylaminopurine(BA) or thidiazuron(TDZ) combined with α-naphthylacetic acid(NAA) was helpful to induce callus.The optimal concentrations of BA were 0.2~0.5mg/L.Nodular stem segments and stem segments were the most suitable explants for callus induction.The medium containing BA and NAA promoted the multiplication of callus.The multiplication rate and differentiation rate of 30-day callus were much higher than those of 60-day callus.BA was helpful to induce callus differentiation.The callus from leaf segments exerted a higher differentiation rate.Conclusion A high-efficiency regeneration system of callus induction and plant regeneration from Pogostemon cablin(Blanco)Benth.has been established.
