长雄野生稻地下茎cDNA文库的构建及EST分析

【摘要】 长雄野生稻(Oryza longisatminata)是广泛生长在热带非洲的1个野生种,具有明显的地下茎特征。为了探索控制其地下茎发育的相关基因及其他优异基因,初步构建了地下茎cDNA文库。文库指标显示:初始滴度为2.5×105cfu/μg,扩增滴度为2.9×108cfu/μg,库容为5×106cfu,重组率为82%;随机挑选80个单克隆进行测序,插入片段在400~1000bp之间;对测序结果进行比对分析,获得了58个已报道有一定功能的EST序列,22个未见报道的EST序列;并利用PAUP4.0软件对序列进行了筛选分析。

【Abstract】 Oryza longisatminata is a wild species which widely grows in tropical Africa, with apparent rhizomatous feature. It is one of the species of the wild rice Oryza rufipogon. To explore the gene which controls the rhizomatous development as well as other favorable genes, a cDNA library was constructed. According to the cDNA library the initial titer of rhizomatous cDNA library was 2.5 ×105 cfu/μg, the amplification titer 2.9×108 cfu/μg, the storage capacity 5×106 cfu, and the recombination rate 82%. Eighty monoclones were selected randomly for sequencing, and most of the insertion fragments ranged from 400 bp to 1 000 bp. Blast analysis of the sequencing results were carried out, and 58 EST sequences were obtained, whose certain functions have been reported, while the other 22 sequences have not been reported. A screening analysis had been done with the PAUP4.0 software, which might provide some references for the cloning and application of related rhizomatous genes of O. longisatminata.
