低温胁迫对草菇菌丝抗氧化酶表达的影响
Effect of Chilling Stress on the Expression of Antioxidative Enzymes in Volvariella volvacea Mycelia
【摘要】 研究低温处理不同时间对草菇菌丝体内超氧化物岐化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)、抗坏血酸过氧化物酶(APX)、脂氧合酶(LOX)和酯酶(EST)等同工酶的影响。结果表明,低温处理2~8h可以不同程度地增强SOD、POD和APX同工酶的表达,并且诱导APX-II新的同工酶表达。SOD、POD和APX活性均呈现先升高后下降的趋势,而CAT活性则随着处理时间的延长逐渐降低。在本试验时段内,随着低温处理时间的延长,草菇菌丝体LOX和EST同工酶表达亦逐渐增强,MDA含量与处理时间呈显著正相关。低温处理不改变草菇菌丝蛋白质含量,但处理8h时菌丝体部分大分子量蛋白被降解为小分子量蛋白。
【Abstract】 Fungus Volvariella volvacea was cultured in a liquid medium under low temperature(at 5 ℃) for different hours from 2~32 hours to observe the activities of isozymes of superoxide dismutase (SOD), peroxidase (POD), catalase(CAT), ascorbate peroxidase(APX), lipoxygenase(LOX) and esterases (EST) in its mycelia. Analysis of native PAGE enzyme activity staining revealed that chilling stress modulated the activities of SOD, POD, CAT, APX, LOX and EST. The activities of the isozymes of POD, SOD and APX in mycelia were enhanced to some extents under cold treatment for 2~8 hours and an additional APX isozyme was induced. The activities of the POD, SOD and APX enzymes all increased initially and then began to decline, but remained higher than that of the control. The activity of CAT was decreased, while the activities of LOX and EST were up -regulated during the whole treatment time. The content of malondialdehyde was positively correlated with treatment time. The content of proteins had little change under cold treatment, but the proteins with high molecular weight in mycelia were decomposed into proteins with low molecular weight at 8 hours after the cold treatment.
