【摘要】 目的研究人乳头瘤病毒16型E6蛋白在COS-7细胞中的定位及表达。方法构建pEGFP-C1介导的HPV-16E6的真核表达载体pGFP-16E6,体外转染COS-7细胞,采用荧光显微镜观察HPV-16E6蛋白在COS-7细胞中的定位及表达。结果细胞在转染pGFP-16E6质粒后,细胞核发出明亮的绿色荧光,GFP-16E6于转染后6h开始表达,至21h表达到高峰,随后表达逐渐降低。结论HPV-16E6蛋白主要表达于COS-7细胞核内,并且表达随时间成动态变化。更多还原

【Abstract】 Objective To observe the expression level and location of HPV-16E6 in COS-7 cells.Methods COS-7 cell was transfected with pGFP-16E6.The fluorescent microscope was used to observe the HPV-16E6 location and expression level.Results After transfection with the pGFP-16E6,there were brilliant fluorescence in the nuclei of COS-7 cells.Furthermore,the GFP-16E6 was expressed from 6h post-transfection,reached the highest level at 21h after transfection and then the expression degraded gradually.Conclusion HPV-16E6 was mainly expressed in nuclei of COS-7 cells.Its expression level was related with time course.更多还原
