重金属离子对猪血SOD的活力与构象影响
Effects of Heavy Metal Ions on the Activity and Conformation of Superoxide Dismutase (SOD) from Porcine Blood
【摘要】 以猪血为材料,以超氧化歧化酶(简称SOD,EC 1.15.1.1)为研究对象,经有机溶剂沉淀法得SOD酶液,测定SOD酶中铜锌含量比例.研究Hg2+、Pb2+、Cd2+等不同重金属离子对SOD活力与构象影响,结果表明:猪血SOD酶中含有等量的铜与锌.Hg2+、Cd2+对酶活力均表现为低浓度激活高浓度抑制作用,而Pb2+在考察的浓度范围内表现激活作用,荧光光谱显示三种重金属离子的存在均引起酶构象的改变.
【Abstract】 Superoxide dismutase(SOD.EC 1.15.1.1) from porcine blood was studied.SOD was purified by precipitation with organic solvent.The contents and function of copper and zinc in SOD,and the effects of heavy metal ions such as Hg2+、Pb2+、Cd2+ on the activity and conformation of enzyme were investigated.The results show that porcine blood SOD contains the same amount of copper and zinc.Hg2+ and Cd2+ will make the enzyme active at lower concentrations,but inhibit the enzyme activity at higher concentration.However,Pb2+ will increase the enzyme activity.Hg2+、Pb2+、Cd2+ will change the enzyme conformation.
