南通市沙门菌毒力基因分布特征的初步研究
The preliminary study about distribution features of Salmonella virulence genes in Nantong city
【摘要】 目的对南通市分离的沙门菌进行毒力基因检测,了解其携带毒力基因的情况,探索毒力基因用于沙门菌分型的可能性。方法运用生物化学、血清凝集等实验对实验菌株进行鉴定,使用多重引物PCR方法对经鉴定的沙门菌进行毒力基因检测。结果11个被检测的毒力基因invA、orgA、prgH、spaN、tolC、sipB、sitC、pagC、msgA、spiA及iroN在所有菌株中都存在;另5个基因,在菌株中的存在情况不同,lpfC和sifA基因在阿贡纳和德尔卑中存在更广泛(80%、90%和73%、77%);cdtB、pefA和spvB基因在实验菌株中的检测阳性率不高,并且只存在于少数几个血清型(埃森和德尔卑)。结论南通市分离的沙门菌携带多种毒力基因,且菌株之间有一定差异,毒力基因用于基因分型有待于进一步探讨。
【Abstract】 Objective To find out how salmonellas separated in Nantong carry virulence genes by practicing gene tests of virulence genes,and explore the possibility of applying the technology to typing salmonellas. Methods Biochemical and serum agglutination tests were used in identifying salmonellas,which were tested by virulence genes with multiple-PCR method. Results Through detection, genes such as invA,orgA,prgH,spaN,tolC,sipB,sitC,pagC,msgA,spiA and iroN existing all strained, which was different in the others. lpfC and sifA occurred wider in S.agona(80%,90% and 73%,77% separately), while cdtB,pefA and spvB had a low positive rate, only in several serotypes.Conclusions Nantong Salmonellas which are separated carry manifold virulence genes which distribute differently in each strain.Applying virulence genes to genotyping is worthy of further study.
