整合素αvβ6亚基αv及β6对卵巢癌细胞系HO-8910PM增殖及侵袭的作用

【摘要】 目的:研究整合素αvβ6亚基αv及β6对高转移卵巢癌细胞系HO-8910PM增殖及侵袭的影响。方法:流式细胞仪检测HO-8910PM细胞αv及β6阳性表达率,MTT实验观察抗αv及抗β6单抗对HO-8910PM细胞增殖的影响,细胞侵袭实验观察抗αv及β6单抗对该细胞系侵袭的影响。结果:HO-8910PM细胞胞质内表达αvβ6,但表达率均不高,αv阳性表达率为25.7%,β6为28.3%;MTT实验显示,αv单抗可明显抑制HO-8910PM细胞增殖(P<0.05),β6单抗抑制肿瘤增殖作用不明显。细胞侵袭实验显示,β6单抗能明显抑制HO-8910PM细胞对人工基底膜胶的侵袭穿透能力(P<0.05)。结论:整合素αvβ6的αv亚基主要参与卵巢癌肿瘤细胞的增殖过程,β6亚基更多参与卵巢癌肿瘤细胞的侵袭过程。

【Abstract】 OBJECTIVE:To investigate the influence of subunits αv and β6 of integrin αvβ6 on proliferation and invasion of ovarian cancer cell HO-8910PM.METHODS:Detect the expression of integrin αvβ6 in cytoplasm of HO-8910PM cells with flow cytometry,and confirm the positive expression of αv and β6 in this cell line.The influence of proliferation in HO-8910PM was observed by MTT test.The influence of invation in HO-8910PM with different concertration and on different time was observed by Transwell chambers.RESULTS:Subunits αv and β6 of integrin αvβ6 were expressed in the cytoplasm of ovarian cancer cell HO-8910PM.But the expression rate is not high.The average positive expression rates of αv and β6 are 25.7% and 28.3%.The proliferation test showed that anti-αv inhibited the cells’ proliferation significantly (P<0.05).Anti-β6 inhibited the cells’ proliferation little.The result of Transwell system showed that anti-β6 inhibited the invasion in the artificial basement membrane significantly (P<0.05).CONCLUSION:Subunit αv may participate more in the process of ovarian cancer cells’ proliferation,and subunit β6 may participate more in the process of ovarian cancer cells’ invasion.
