骨肉瘤组织FLIP和FADD及Caspase-8蛋白表达与细胞凋亡相关性的研究

【摘要】 目的:探讨FLIP、FADD和Caspase-8在人骨肉瘤组织及骨软骨瘤组织中的表达及其与骨肉瘤细胞凋亡的关系。方法:应用免疫组化法检测FLIP、FADD和Caspase-8蛋白在43例骨肉瘤组织及30例骨软骨瘤组织中的表达,并经图像分析系统检测FLIP、FADD和Caspase-8蛋白的平均光密度(OD)值。用激光扫描共聚焦显微镜检测骨肉瘤及骨软骨瘤中细胞凋亡分布情况并对各因子之间进行相关性分析。结果:FLIP蛋白阳性物质在骨肉瘤中的表达高于骨软骨瘤,P<0.05;而FADD和Caspase-8蛋白在骨肉瘤中的表达低于骨软骨瘤,P<0.05。FLIP和Caspase-8蛋白在骨肉瘤中表达呈负相关,r=-0.843,P<0.05;而FLIP、FADD蛋白在骨肉瘤中表达无相关性。骨肉瘤组细胞凋亡指数(AI)显著低于骨软骨瘤组,P<0.05;FLIP蛋白表达与细胞凋亡指数呈负相关,r=-0.726,P<0.05;而Caspase-8蛋白表达与骨肉瘤细胞凋亡呈正相关,r=0.446,P<0.05;FADD蛋白表达与与凋亡指数无相关性。结论:FLIP蛋白对骨肉瘤细胞凋亡有抑制作用,Caspase-8蛋白在骨肉瘤中呈低表达,抑制了肿瘤细胞凋亡。如能抑制FLIP的表达,促进Caspase-8的表达,可提高骨肉瘤细胞对凋亡的敏感性。

【Abstract】 OBJECTIVE: To investigate the expressions and distributions of FLIP,FADD and Caspase-8,as well as the relationship between their expressions and apoptosis in human osteosarcoma.METHODS: Immunohistochemistry was used to detect the distributions of FLIP,FADD and Caspase-8 proteins by measureing the average optical density with image analysis system for 43 patients with osteosarcomas and 30 patients with osteochondroma.The apoptosis was detected by TUNEL,and the apoptosis cells could be seen with the fluorescence-FITC labeling method by a confocal laser scanning microscope.The correlated analysis was performect among FLIP,FADD,Caspase-8,and apoptosis.RESULTS: The positive expression of FLIP protein in osteosarcoma was significantly higher than that in osteochondroma(P<0.05),while the positive expressions of FADD and Caspase-8 proteins were significantly lower than those in the osteochondroma group(P<0.05).There was a negative correlation between the FLIP and Caspase-8 protein expressions(r=-0.843,P<0.05).Whereas,there was no correlation between FLIP and FADD protein expressions in osteosarcoma.The apoptosis index(AI) was lower in osteosarcoma than that in the osteochondroma group(P<0.05).There was a negative correlation between the FLIP protein and apoptosis(r=-0.726,P<0.05),and a positive correlation between Caspase-8 protein and apoptosis was found(r=0.446,P<0.05).There was no correlation between FADD protein and apoptosis.CONCLUSIONS:It is suggested that FLIP protein can inhibit apoptosis of osteosarcoma.The lower expression of Caspase-8 can restrain apoptosis in osteosarcoma.Depress expression of FLIP and inspire expression of Caspase-8 may promote the sensitvity of apoptosis effect of osteosarcoma.
