血管抑素基因联合5-FU腹腔化疗治疗大肠癌术后腹腔种植的实验研究

【摘要】 目的:观察血管抑素基因联合5-氟尿嘧啶(5-FU)腹腔化疗对预防大肠癌术后腹腔种植的作用。方法:RT-PCR扩增血管抑素基因,构建重组腺病毒载体。建立人结肠癌LoVo细胞腹腔种植的模型,给予血管抑素基因联合5-FU腹腔化疗,并观察其对大肠癌细胞腹腔种植的影响。结果:各组裸鼠的致瘤率和死亡率为100%。血管抑素基因联合5-FU腹腔化疗治疗组的腹水出现时间晚〔(22.3±5.4)d〕,荷瘤生存时间长〔(47.3±8.4)d〕,治疗组肿瘤组织中血管抑素基因表达为阳性,肿瘤微血管密度(3.64±0.58)明显低于其他各组。结论:血管抑素的基因治疗与传统的腹腔化疗联合应用,可能是预防大肠癌术后腹腔种植的一种有效方法。

【Abstract】 OBJECTIVE:To study the thereopeutic effect of combination of angiostatin gene therapy and peritoneal chemotherapy with 5-FU to peritoneal implantation of colonic carcinoma.METHODS: Angiostatin gene was cloned into adenovirus vector.LoVo cells inplanting model was built,which were treated with combining angiostatin gene therapy with peritoneal chemotherapy with 5-FU or other therapy.RESULTS:Both the rates of sickness and the rate of death were 100%.The survival time in the group of the combination of angiostatin gene therapy and peritoneal chemotherapy with 5-FU((47.3±8.4) d) was much longer than that in other groups.And ascites was found late((22.3±5.4) d).There were much less microvessels(3.64±0.58)and stronger angiostatin gene expression in tumor tissues in the treatment group.CONCLUSION: It may be a good method to treat peritoneal implantation of colonic carcinoma with angiostatin gene therapy combinined with peritoneal chemotherapy with 5-FU.
